  Preeclampsia is a pregnancy-specific disorder associated with hyperlipidemia. Liver X receptor (LXR) alpha and LXRbeta are key regulators of lipid homeostasis. In the current study, we investigated expression of LXRalpha, LXRbeta and their target genes in human term placenta, decidua and subcutaneous adipose tissue from pregnancies complicated by preeclampsia. Furthermore, we analyzed the protein levels of LXRalpha and LXRbeta in placenta. We also analyzed lipid concentrations in term placental tissue. Gene expression of LXRalpha, LXRbeta and fatty acid transporter CD36 was significantly decreased in placental tissues, while increased expression was observed for LXRalpha in adipose tissue, from pregnancies complicated by preeclampsia. The placental protein level of LXRbeta was reduced, and there was a positive correlation between placental LXRbeta mRNA expression and placental free fatty acids in preeclampsia. Our results suggest a possible role for LXRbeta as a transcriptional regulator in preeclampsia.

  Fruits and vegetables have been thought to be beneficial in cardiovascular disease. The beneficial effects of fruits and vegetables may be explained by the antioxidants and other components contained therein. These nutrients may function individually or in concert to protect lipoproteins and vascular cells from oxidation, or by other mechanisms such as reducing plasma lipid levels (LDL cholesterol, triglycerides), and platelet aggregation response. Kiwi fruit which contains high amounts of vitamin C, vitamin E and polyphenols may be beneficial in cardiovascular disease, however very little is known about its cardioprotective effects. Platelets are involved in atherosclerotic disease development and the reduction of platelet activity by medications reduces the incidence and severity of disease. To this end, we evaluated whether consuming kiwi fruits modulated platelet activity and plasma lipids in human volunteers in a randomized cross-over study. We report that consuming 2 or 3 kiwi fruits per day for 28 days reduced platelet aggregation response to collagen and ADP by 18% compared with the controls (p<0.05). In addition, consumption of kiwi fruits lowered blood triglycerides levels by 15% compared with control (p<0.05) whereas no such effects were observed in the case of cholesterol levels. All these data indicate that consuming kiwi fruits may be beneficial in cardiovascular disease.

  In the feto-placental unit, preferential transport of maternal plasma arachidonic acid (20:4n-6) and docosahexaenoic acid (22:5n-3) across the placenta is of critical importance for fetal growth and development. More than 90 per cent of the fat deposition in the fetus occurs in the last 10 weeks of pregnancy. All of the n -3 and n -6 fatty acid structure acquired by the fetus has to cross the placenta and fetal blood is enriched in long chain polyunsaturated fatty acids (LCPUFA) relative to the maternal supply. Fatty acids cross the microvillous and basal membranes by simple diffusion and via the action of membrane bound (FAT, FATP and p-FABPpm) and cytoplasmic fatty acid-binding proteins (FABPs). The direction and magnitude of fatty acid flux is mainly dictated by the relative abundance of available binding sites. The existence of a fatty-acid-transport system comprising multiple binding proteins in human placenta may be essential to facilitate the preferential transport of maternal plasma fatty acids in order to meet the requirements of the growing fetus. The critical importance of long-chain fatty acids in cellular homeostasis demands an efficient uptake system for these fatty acids and their metabolism in tissues. In fact, involvement of several nuclear transcription factors (PPAR&#61543;, LXR, RXR, and SREBP-1) is critical in the expression of genes responsible for fatty acids uptake, placental trophoblast differentiation and hCG production. These indicate that these receptors are potential regulators of placental lipid transfer and homeostasis. This review discusses importance of nuclear receptors and fatty acid binding/transport proteins in placental fatty acid uptake, transport and metabolism.

  The effects of fatty acids on acyl-CoA thioesterase activity and peroxisome proliferator-activated receptor gamma (PPARgamma), a regulator of lipid metabolism, were investigated in placental choriocarcinoma (BeWo) cells. Substrate preference for acyl-CoA thioesterase was in the following order; gamma-linolenoyol-CoA>/=arachidonoyol-CoAz.Gt;palmitoyl-CoA>/=linoleyol-CoA. However, when these cells were incubated with fatty acids, acyl-CoA thioesterase activity was increased by both conjugated linoleic and gamma linolenic acids, but not by docosahexaenoic and eicosapentaenoic acids. In addition, these fatty acids also increased expression of PPARgamma in these cells, suggesting a putative relationship between free fatty acid generated by acyl-CoA thioesterase and expression of PPARgamma. Since expression of PPARgamma is critical for feto-placental growth, these fatty acids may be important during pregnancy.

  The strong positive relationship between cholesterol and coronary heart disease (CHD) can be derived from several lines of evidence [1,2]. Significant reduction in the incidence of clinical coronary events has been shown even with relatively modest changes in low-density lipoprotein cholesterol (LDL-C) using various lipid-lowering drug regimens [3]. However, the improvement in cardiovascular end points is incompletely explained by the treated LDL cholesterol levels, and other mechanisms of action of lipid-lowering drugs, particularly the statins (3-hydroxy-3-methylglutaryl reductase inhibitors) have been recognised. These include modification of endothelial function, inflammatory responses, effects on plaque stability and thrombus formation

  Arachidonic acid at 100nM stimulated internalisation of 125I-leptin in human placental choriocarcinoma (BeWo) cells by 3-fold compared with controls. In contrast, eicosapentaenoic acid at similar concentration decreased internalisation of leptin by 2-fold. Use of ibuprofen and indomethacin (inhibitors of prostaglandin synthesis) inhibited the stimulatory effect of arachidonic acid. Prostaglandin E(2), a cyclooxygenase metabolite of arachidonic acid, stimulated internalisation of leptin by these cells. All these data demonstrate that stimulation of leptin internalisation by arachidonic acid in placental trophoblasts may be mediated via prostaglandin E(2).

  Increased platelet reactivity has been implicated in the vascular complications of myeloproliferative diseases and diabetes mellitus. The mechanisms of platelet hyperresponsiveness have not been fully explained. Expression of CD36 or fatty acid translocase (FAT) and its role in arachidonic acid (AA) uptake by platelets were examined in subjects with myeloproliferative disorders(MPD), those with non-insulin-dependent diabetes mellitus (NIDDM), and in normal, healthy, age-matched controls. Surface expression of CD36 on platelet membranes was increased in MPD (10.94¡Ó0.76 pmol/mg protein) compared with normal controls (6.94¡Ó0.48 pmol/mg protein), p¿¬0.001. Total platelet content of CD36 was also significantly higher (32.1¿b0.61 pmol/mg protein, p<0.01) compared with those in sex and age matched normal controls (25.7¿b1.09 pmol/mg protein). In contrast, platelet surface expression of CD36 in NIDDM (6.5 ¿b 0.56 pmol/mg protein) was not significantly different from those of normal controls despite higher total content of CD36 (32.8¿b1.2, pmol/mg protein p<0.01). Intact MPD platelets bound significantly more arachidonic acid (AA) (1.53 ¡Ó0.16 nmol/mg protein, p<0.05), compared with controls (1.12¡Ó0.07 nmol/mg protein) or NIDDM subjects (1.16¿b0.16 nmol/mg protein). The capacity of MPD platelet membranes to bind 14C-AA was also increased (1.72¡Ó0.25 nmol/ protein, (p<0.05) compared with that of controls (1.62 ¡Ó 0.05 nmol/protein) and of NIDDM (1.22 ¿b 0.08 nmol/protein). This is consistent with higher surface expression of CD36 in MPD platelets. Membrane fatty acid analysis indicated that the % of AA in platelet phospholipids was significantly lower in MPD (3.15 ¡Ó 0.81%) compared with the controls (5.62 ¡Ó 1.72 %), p<0.05. The AA content of diabetic platelets (4.82¿b1.15%) was not significantly different from normal controls. In summary, both total and surface expression of CD36 are increased in MPD, consistent with an enhanced capacity for uptake of AA by platelets. Increased expression of CD36 in platelets may play a role in the vaso-occlusive manifestations of MPD.
