DNA cloning

Cloning of vectors for random insertion or homologous recombination is simplified by using proven techniques and material.

Introduction and considerations

NTS may assist with cloning of regular expression vectors, targeting vectors (for homologous recombination), and transgenic constructs (for random or site-specific integration). Cloning of vectors for random insertion will be based on PCR amplification, restriction digestion and ligation. Targeting vectors typically starts with a BAC clone, extraction of a desired gene segment from the BAC clone into a suitable vector, followed by insertion of desired elements (e.g. LoxP sites and selection cassette) with recombineering technology.

Description of the service

  • NTS has a long-lasting experience in molecular cloning techniques (regular restriction enzyme/ligation, recombineering, BAC engineering, CRISPR-Cas9).
  • NTS has a stock of useful vector components that can be used.

Technical service performed by NTS

  • NTS may assist with cloning of the construct.
  • Whenever needed, NTS also purchases reagents that will simplify or improve cloning of a given construct.

Costs cover the following

  • The cost for the purchase of reagents will be charged the customer.

Conditions

  • Materials submitted by the customer to NTS will remain the property of the customer and will not be used for other projects unless such permit has been given by the customer.
  • Depending on the nature of the work involved, this service may be considered as scientific input to the research project. Such projects will be performed under agreement that one or two persons from NTS will be co-authored on the first publication.
