Project summary

Interstitial lung disease (ILD) refers to a group of conditions characterized by lung fibrosis, including idiopathic pulmonary fibrosis (IPF), a fatal disease with a median survival of ~3 years. Current treatments, Pirfenidone and Nintedanib, have limited efficacy and significant side effects, underscoring the urgent need for better therapeutic options.

Researchers at Oslo University Hospital are advancing a novel anti-fibrotic treatment using OM-153, a highly potent tankyrase inhibitor targeting TNKS/WNT/YAP signalling. Early studies demonstrate promising anti-fibrotic effects in cell culture, human patient models, and animal models. The project focuses on developing and validating advanced fibrosis research models that utilize multi-component stimuli instead of traditional single TGFβ-based approaches to induce fibrogenesis.

These cutting-edge models, combined with state-of-the-art lung fibrosis research techniques, will provide deeper insights into the therapeutic effects and mechanisms of TNKS inhibitor treatments.

TNKS inhibition counteract pro-fibrotic WNT/YAP signaling pathways activated by inflammatory cytokines, and consequently fibroblast-myofibroblast transition (FMT) and fibrogenesis/extracellular matrix (ECM) deposition.
