Forskning

Bjarne Bogen forskning har vært fokuser på T og B celler og deres betydning for autoimmune sykdommer og immunovervåkning av kreft. Han har i den senere tid også arbeidet med utvikling av nye og bedre vaksiner. Nedenfor er en kort oversikt over disse studiene, hvor mange medarbeidere over mange år har gitt betydelige bidrag (se liste over originale forskningsrapporter og oversikt over forskningsgruppen "Cellulær og molekylær immunologi").

1. Idiotype-drevet T-B celle samarbeid, autoimmune sykdommer og utvikling av kreft i B celler (B celle lymfomer). Idiotoper (Id) er antigene determinanter som er lokalisert til immunglobulin (Ig) variable (V) regioner. Bogen beskrev at B celler nedbryter deres Ig reseptor (BCR) for antigen, og at fragmenter av V regionene blir presentert på MHC ("Major Histocompatibility Complex") klasse II molekyler til Id-spesifikke CD4+ T celler. Dette resulterer i Id-drevet T-B celle samarbeid. I videre arbeider viste han at Id-drevet T-B samarbeid kan resultere iutvikling av  autoimmune sykdommer og  B celle lymfomer (PNAS -89, EMBO-J -89, Cell- 91, EMBO-J -93, EJI -99, JI -05,JI -07, JEM -07, Am J Pathol -09).

2. Immunovervåkning av kreft mediert av CD4+ T celler. Bogen viste at Id-spesifikke T celle reseptor transgene mus er beskyttet mot Id+ B celle lymfomer og myelomatose. Han viste videre at monoklonalt Ig utskilt av kreftcelelr blir tatt opp av dendrittiske celler og makrofager som infiltrerer svulstvevet. De sistnevnte cellene nedbryter delvis det monoklonale Ig og presenterer Id-peptider på sine MHC II molekyler til Id-spesifikke CD4+ T celler som har migrert til svulsten etter først å ha blitt stimulert i drenerende lymfeknuter. De svulstinfiltrerende T cellene produserer IFNg som aktiverer makrofager slik at de blir cytotoksiske og dreper kreftceller (PNAS -94, PNAS 2000, Blood -01, Blood -03, Immunity -05, Cancer Res -09, Nat commun -11).

3. Nye vaksine molekyler: Troybodies og Vaccibodies. Troybodies er Ig-baserte  molekyler som har T celle epitoper innskutt i løkker mellom beta "strenger" i konstante domener av molekylet. I tillegg er Troybodies utstyrt med nye V regioner med spesifisitet for antigen-presenterende celler (APC). Dette resulterer i forsterket presentasjon av T celle epitoper på APC  MHC molekyler , og dermed økte T celle responser (Nat biotech -99, PNAS -01, JI -02, JI -03, EJI -05). Vaccibodies er Ig-baserte dimere molekyler som har integrert store antigener med intakte B celle determinanter. Vaccibodies målstyres til antigenpresenterende celler enten ved bruk av naturlige ligander (f. eks. kjemokiner) eller scFv spesifikke for APC. Vaccibodies øker både antistoff og T celle responser. Vaccibodies kan leveres som DNA vaksiner. For tiden utvikles vaccibodies for en rekke forskjellige infeksjonssykdommer slik som influenza, HIV, tuberkulose, og visse former for kreft (myelomatose, B celle lymfomer, maligne melanomer).
